Intraperitoneal injection of 2-DG induces Fos expression in the hypothalamus supraoptic nucleus and paraventricular nucleus of rats

Abstract:

Objective To study if intraperitoneal injection of 2-deoxy-D-glucose (2-DG) can functionally activate neurons in the hypothalamus supraoptic nucleus (SON) and paraventricular nucleus (PVN) by induction of Fos expressing.
Methods Twelve healthy male Sprague-Dawley rats were randomly divided into intraperitoneal injection of 2-DG (6 rats), saline control (3 rats) and normal control (3 rats) groups. After the treatment, immunohistochemical staining was employed to observe the expressions of Fos and double labelling of oxytocin (OT) or vasopressin (VP) with Fos in SON and PVN, respectively. The plasmal level of OT and VP was examined by ELISA assay. Results Compared with saline and normal control groups, 2-DG induced specific Fos expression was mainly located in lateral and perifornical areas of hypothalamus. High levels of Fos immunoreactivity were observed in SON and PVN where they double labeled with the specific neuroactive substances OT and VP. The double labeling rates for OT and Fos (the percentage between the number of OT/Fos doublelabeled neurons to that of OT positive neurons) were 87.10% in SON and 90.57% in PVN, significantly higher than that of VP/Fos double labeling rate (68.42% in SON, 76.92% in PVN, P<0.05). In contrast, the plasma level of OT and VP after injection of 2-DG may not show discernible changes to that of control rat. Conclusion Intraperitoneal injection of 2-DG can induce Fos expression in OT and VP neurons in hypothalamic SON and PVN which may be involved in triggering acute stress response induced by 2-DG.
