Protective effects of ethyl acetate extract from Chroogomphus rutilus on the degeneration and death of dopaminergic neurons induced by lipopolysaccharides

Abstract:

Objective To investigate the protective effects of different doses of Ethyl Acetate (EA) Extract from Chroogomphus rutilus on the degeneration and death of dopaminergic neurons of the PD mouse which was induced by lipopolysaccharides (LPS). Methods Adult male Kunming(KM) mice were used in the present study, LPS was injected into the substantia nigra compacta(SNc) to duplicate mouse model of PD. A tatal of 30 KM mice were randomly divided into five groups: the sham group, the LPS control group, low dose experimental group(LPS+100mg/kg), medial dose experimental group (LPS+200mg/kg) and high dose experimental group(LPS+400mg/kg), six mice per group. The immunofluorescence methods were used to label the dopaminergic neurons and astrocytes in SN. The dopaminergic positive neurons were counted and analyzed with statistics. Results The dopaminergic neuron numbers in LPS control group were markedly decreased in comparison with sham group (P<0.05), and the dopaminergic neurons appeared ovale, and the neurite was shorter than the sham group. Astrocytes displayed the activated state with larger bodies, thickened and shortened enation in the SN of LPS control group compared with the sham group. The dopaminergic neurons were increased significantly in the low and median dose groups compared with LPS control group(P<0.05), the density of astrocytes was lower. There was no significant difference of dopaminergic neurons between high dose group and LPS control group(P>0.05). Conclusion The low and median groups may protect the degeneration of dopaminergic neurons in SNc by depressing the activity of astrocytes in SNc.
