Effects of transient focal cerebral ischemia on proliferation of neuronal precursors in adult rats subgranular zone

Abstract:

Objective To investigate proliferation changes of neuronal precursors in subgranular zone (SGZ) of adult rats after transient focal cerebral ischemia.
Methods Ischemia was induced by occlusion of the middle cerebral artery in rats for 90min. The rats were given intraperitoneal injection of BrdU on the 1st and 2nd day after injury, and then were sacrificed on the 3rd, 7th, 14th and 28th day respectively, 6 rats in time point. The brains were removed and frozen sections were cut. Immunofluorescent staining was applied to observe the expression of Brdu, DCX, ki67, GFAP and CNP in SGZ. Results (1)The Brdu(+) cells assembled into clusters. The number of clusters increased at 3d and reached to peak at 7d,and then decreased gradually.(2)The cells in clusters increased at 3d and reached to peak at 7d, and then decreased gradually.(3)The rate of ki67(+) cells decreased from 3d to 28d. Conclusion MCAO/R induces proliferation of neuronal precursors in SGZ. Some of the progeny cells retain their ability to proliferate, and the others differentiate into neurons.
