Abstract:

bjective To investigate the role of tetrahydroxy stilbene glucoside(TSG) on histone deacetylase 1 (HDAC1) level in cerebral ischemic-reperfusion (I/R) injury in gerbils. Methods Sixty-four gerbils were randomly divided into four groups: sham group, model control group, TSG treatment group and the positive control medicine of edaravone injection group. The functions of learning and memory were detected by Morris water maze after 7 days. The cerebral infarction of protective action was investigated by nerve function score and triphenyltetrazolium chloride (TTC) staining. The protein and mRNA level of HDAC1 were detected by the immunohistochemical SP, Western blotting and RT-PCR methods. Results Compared with the model group, TSG had protective effect on ischemia reperfusion of the brain. TSG significantly enhanced the ability of learning and memory in I/R injury model gerbils. The protein expression and mRNA level of HDAC1 were significantly increased after TSG treatment in I/R injury model mice. Conclusion TSG has positively protective and therapeutic effect on cerebral I/R injury in gerbils. Its mechanism may be relevant with maintaining the homeostasis of histone acetylation and deacetylation.
