Influence of galectin-3 low expression on proliferation of MGC-803 cells

Abstract:

Objective To investigate the influence of galectin-3 suppression on the proliferation of human stomach carcinoma MGC-803 cells by down-regulating galectin-3 expression using RNA interference technique. Methods The MGC-803 cells were divided into three groups. The cells transfected with siRNA targeted galectin-3 was regarded as si-RNA interference group, the cells transfected with scrambled siRNA was taken for negative control and those with lipofectine TM2000 alone was blank control. Western blotting was applied to detect the suppression rate of galectin-3. The growth of the cells was assessed by MTT and colony formation assays. Western blotting and immunohistochemistry were used to detect the expression of cyclin D1 and proliferationg cell nuclear antigen(PCNA). Results The expression of galectin-3 was significantly inhibited in the experiment cells group when compared with those of the blank cells group and negative cells group (P<0.05). MTT and colony formation assays showed that proliferation of the experiment cells group decreased significantly (P<0.05). The expression of cyclin D1 and PCNA decreased compared with the negative control group and the blank control group (P<0.05). Conclusion Galectin-3 low expression inhibits the proliferation of MGC-803 cells which might associate with the suppression of cyclin D1 and PCNA expression.
